From the live streams and videos on YouTube I've seen: no, you can't use the buttons in the main UI, or in some game menus. It appears to be a bit annoying, as I would prefer the choice, but at least the menu isn't completely terrible to navigate with touch (or doesn't appear to be).

A lot of the screens on the Vita don't let you use the buttons, especially the primary UI, according to many of the videos and live streams going around. They force you to use the touch screen only, and there isn't a setting to change it.

I'm actually somewhat fascinated by this...not all 'DO WANT!' but definitely curious.
