Best surprise? South Park: The Stick of Truth. I thought it would be funny, but I didn't expect it to be nearly as funny (or consistent) as it was. It had Mario RPG-style gameplay that didn't get old and a wonderful pace that never felt like it was killing time.
Mass Effect 2 delivered a couple fantastic ones. I mentioned Mordin singing "Scientist Salarian" as a beautiful and unexpected moment in a reply to someone below. My favorite, though, has to be this:
Mine is also a Mordin moment, but of a completely different nature. My favorite moment of sheer, unexpected joy in a game.
