I'd disagree. Strictly going off of a my last viewing of this movie from about a decade ago - but I recall a good chunk of the movie devoted to Cary Grant's character deciding to annul the marriage as he thought he had crazy genes and the fallout that ensued. I'd say that counts as romantic subplot.

Romantic comedies were not "born" with Annie Hall. The genre preceded AH by decades. Hell, look at Cary Grant's filmography - Arsenic and Old Lace; His Girl Friday; Charade; Bringing Up Baby - these are all romantic comedies, and that's just one leading man's resume.
