You don't need to be a gamer to notice the effects of fragmentation, which are very little. The biggest speed boost would be to get a SSD.

From what I've read though, it doesn't really "sandbox" any app. It just packages an app in the paf format for easy integration in the PortableApps launcher.

If you have some spare money laying around, I would suggest switching the HDD out for an SSD. With an SSD, waiting time for apps to load will be greatly reduced, and everything will feel much snappier. Also, because SSDs have no movable parts, it will also increase the battery life.
