Technically only three bosses per wing, but there's also Heroic mode for each and 2 class challenges per wing where you have to defeat a boss with a pre-built deck.

It is random, but if the only other minion on your side of the field has a Battlecry or a damaged, shield-less Sunwalker, that's still a beneficial effect.
It has been confirmed that a connection will be required for single player, and that is unlikely to ever change; given the game's heavy emphasis on PvP and spending gold/money on packs, allowing the game to store your decks/card collection locally would allow for too much hacking/cheating.
