The conditioning coil should be piped in parallel so closing that valve *shouldn't cause any problems. The boiler should operate on it's own thermostat keeping the water at a set temp, regardless of whether the zones are needing heat or not. Several good answers here, but there should be a manual cutoff valve too. You may want to just turn that off to stop the hot water flowing to that coil. I'm not sure how your zones are setup exactly, but normally there is a valve to control the chilled/heated water to the coil and or a fan control. Maybe the valve
