Do you mean the Eastman comics or the kids television show? Eastman comics were pretty mature, and the Nick TMNT show (despite having great fight scenes) is still a kids show (no real blood is shown).

It doesn't have to have a big fanbase in the beginning to become a hit. Look at the Matrix, it had no fanbase as it was an original IP, and yet look how successful it was. Guardians of the Galaxy was a very much less known comic, but that doesn't take away from it being a good story and even better movie.
