Are you using Simi? If you try this with regular icons, there is default padding around each one I have yet to see circumvented.
The icons are actually 3x3 on the grid, not 1x1. To my knowledge it is impossible to make app icons or stock android folders larger than 1x1. Even if you could, I imagine they wouldn't look very good (even when I tried to use these icons as 1x1 at 135% size with nova, they looked fuzzy and pixelated). That's why I…
Simi has only one way of displaying its apps but you can play with the way that looks. For example; go to launcher settings in a given folder, set launcher style to custom, and set the background color to solid black, dark grey, or even the highlight blue color on ICS/JB. Instantly more Holo themed!

You don't actually have to install the icon pack, Simi allows for custom images to represent its folders. Download and unzip the pack, pick your favorite icons, and put them on storage on your phone. You can select any image on your phone for the folder in Simi-> Folder Setting -> Widget Settings-> Icon
What Asskicker said, making the stock android folders larger looks pixelated and has spacing issues. By using widgets, I can easily make sure the folders fit together nicely.
