One of the Flash or Arrow producers called Supergirl's verse Earth-CBS, which I found amusing.
We saw a couple flashbacks with Earth-1 Wells before he got killed where he was with his wife. He seemed like a really good guy.
Even without the spoiler thing, I don't think they'd go there. M'gann in the comics is a teenager, most often associated with the Teen Titans. Obviously, she's aged up here, but that would be really weird.
Something similar with Batwoman. Kate Kane's backstory has her kicked out of the military academy, because of Don't Ask, Don't Tell. We haven't reached the point where it doesn't make sense quite yet, but eventually she's going to need a new backstory.

In the comics, Yo-Yo is her given name (Yo-Yo Rodriguez). Her code name is Slingshot, so they could change it if they want to.
Hawkgirl was hugely popular in the Justice League Unlimited cartoon. She has little in common with the LoT Hawkgirl though. JLU used the alien cop backstory for her instead of the reincarnated Egyptian princess one.

Cadmus was mentioned back when the DEO first discovered Hank was really J'onn. Cadmus was where they were taking him to be experimented on before Supergirl saved him.
TV Flash is allowed to exist separately from movie Flash because he's been grandfathered in. The show's already on and super popular, so there's not much they can do to change that.
I was really hoping Snart was going to get a cool cybernetic hand.
