You unlock characters as you progress. You play as the Guardians of the Galaxy during the first chapter, then most of the big name characters (Iron Man, Cap, Wolverine, etc.) are added midway through chapter 2. From there, you usually unlock 2-3 characters directly related to the chapter as you progress.
