It was pretty clear in the series that there were a group of 500 people from all the world that, 500 years ago, discovered something or was subject to some experience that made them immortals. Maybe aliens were involved but the immortals themselves were just human beings made immortals.
The original plan of having a few flashbacks of Nikki as a spy only to reveal later that it was a TV show would have been pure genius and pitch perfect for Lost.
