I am very partial to the awkward orgy sketch personally. Also there should be a special award given to Metta World News for best recurring sketch. I can never get enough Metta nonsense.
So glad Matt mentioned Kentucky Route Zero. It's easily one of the most beautiful games I have ever played. The museum set piece in Act II had me in tears, I'm still not sure why. Also gave me one of my favorite pieces of music, "Long Journey Home" by The Bedquilt Ramblers.
