Even though the Screw Attack is the defacto icon for Metroid, I think the Ice Beam does a better job representing the series. It's a weapon and a tool, and it changes both the combat and the exploration dynamics. That's sort of the cornerstone of Metroidvania gameplay: a disguised "key" to the game's "locks."
