I don't doubt that people play the game, but the popularity of the game is by far more relevant to the surge in popularity of lets plays.
The vast majority of the audience for that game will have found it through lets plays, or through the proliferation of information gained form them.

I've never heard anybody english say "slendermun", always "Slender-Man", but on numerous occasions I've heard Americans kind of affix the words into more of a "slendrmn" sound.

That's true, but a good 90% of slender fans only really know of the character from some of the media created featuring him.
