I’d say it’s the other way round - if you only have a single master & apprentice then the apprentice has no choice but to kill the master. Whereas if you have a bunch of masters each with a dozen apprentices then it’s obvious that you can become a master without having to kill for it. Or at least not having to kill you…

From the Wookiepedia article I’d regard the Rule of Two as being put in place to keep the Sith secret (rather than being some major ideological constraint). Once Palpatine was thoroughly in charge, he may not have been that bothered by secrets any more.
Star Wars Rebels also has Maul coming to kill Obi Wan, though Owen doesn’t get involved.
